    The answer should be: yes.

    You need to upgrade the schema to Server 2016. When you install your first 2016 domain controller, the promotion wizard will automatically upgrade your forest schema.

    We are dealing with several different requirements here.

    1. AD FS requires Domain controllers running Windows Server 2008 or later.
    2. New installations of AD FS 2016 require the Active Directory 2016 schema (minimum version 85).
    3. To prepare the Schema for Device Registration, your Active Directory forest must have the Windows Server 2012 R2 schema.

    What information do we have?
    1. All domain controllers run Windows Server 2012 R2.
    – This means our schema is ready to be prepared for Device Registration using the Initialize-ADDeviceRegistration command. It also means we can deploy AD FS in general.
    2. You plan to deploy an AD FS farm on Server1.
    – So this appears to be a new installation and will require Server 2016 schema.

    Conclusion, upgrading a domain controller to Server 2016 will upgrade the schema, meeting all requirements. To me, the answer is YES.

    The answer is no.

    By upgrading A domain controller, it does NOT automatically upgrade the Forest and domain mode with it, as long as there are other domain controllers not running the required version.
    AD FS Device registration needs at least forest mode 2012 to function.

    The devil is in the details with this question.
